Is NAD+ the "fountain of youth"

Aging is definitely not for me….so I’m using NAD+ as my new  found “fountain of youth.”. I don’t like aging any more than the next guy and recently I’ve been noticing little reminders everywhere:  not keeping up with the 20 year olds playing basketball at the gym, the soreness after a workout that lasts 3-4 days now, the fatigue that sets in earlier and earlier in the day and that irritable mood that comes more often now and colors everything in my life pessimistic.

If it’s aging, then I don’t like it.

Surely science has an answer to that by now right?

NAD+: the aging antidote.

Science’s answer might be this “new” thing called NAD+.

(Well, it’s not actually “new” it’s just been rediscovered and finally made available)

A relative of the B3 vitamin Niacin, NAD+ is a co-enzyme found in every living cell. It hangs out in the mitochondria, the little factories that make energy in all of our cells. NAD+ shuttles electrons back and forth and helps convert energy into it’s usable form: ATP.

To help understand how critical this is, think of your cells as cities. For your city to keep things going – schools, police, fire department, new buildings, etc. – it needs money in the form of US dollars. Without money, everything stops.

It’s the same in cells, except instead of US dollars, the currency that cells use is ATP. You need to constantly make new neurotransmitters, hormones, muscle cells, collagen, etc. for your cells to function and without ATP everything slows down to a halt.

As we age, mitochondria  become less efficient and they produce less ATP. A big part of the reason this happens is that the NAD+ inside of mitochondria gets depleted.

So why haven’t we all been taking NAD+ with our breakfast cereal every morning if that’s the case?

Well, as it turns out NAD+ is a finicky molecule and it is very hard to keep it stable in it’s purified “active” (reduced) form.  Most products out there are in it’s inactive (oxidized) form as NADH, but you’ll have a very hard time finding a pure oral NAD+ oral supplement.  Many manufacturers sell NADH while others try to raise NAD+ levels indirectly by using supplements like “Nicotinamide Riboside” or “Niacinamide”. All well and good, and there is some science that show these indirect methods might work to some extent,  but how can you get  PURE REDUCED NAD+ DIRECTLY into your body ?

My first NAD+ IV Infusion

Not long ago, before introducing NAD+ as an treatment at our IV infusion clinic in Alexandria, Virginia, I tried it for myself. I had to see what all the fuss was about.

The nicotinamide adenosine dinucleotide (NAD) molecule alternates between two forms. Which one is present depends on how it is being utilized. NAD+ is the reduced (active) form of the molecule. NADH is the oxidized (inactive) form of the same molecule.

Energy production in your mitochondria—the little energy factories that pump out ATP—ABSOLUTELY depends on NADH (the inactive form) being recycled back to active NAD+. If this recycling stops and NADH accumulates the cell runs out of energy and it may die. This was clearly shown in a recent study on brain and kidney cells.1

It goes without saying, then, that depleted levels of NAD+ and accumulation of NADH, or a low ratio of NAD+/NADH, can create problems for your mitochondria and your cells. Indeed, it results in low levels of cellular energy and may be one of the primary contributing factors to “mitochondrial dysfunction”. a condition we now know is implicated in a wide variety of chronic illnesses including autoimmune disorders, diabetes, and others.

Restoring NAD+ levels reverses this process and allows cells to return to full energy status, offering a possible way to undo mitochondrial damage and the resulting chronic illness.

NAD+ as an Anti-Aging Molecule

Medical researchers have long known that a group of enzymes called sirutins play a crucial role in how the body ages—especially SIRT1 and SIRT3. We don’t yet understand precisely how they work, but what we do know is exciting. SIRT enzymes appear to switch off genes that promote aging such as those that cause inflammation, fat synthesis and storage, and blood sugar management issues.

Up until now the only way we knew to positively impact these SIRT enzymes was to go on a very low-calorie diet. As uncomfortable as it may be, caloric restriction is one of the few ways that has repeatedly been shown to lead to a longer life.

However, the tide appears to be turning. You see, recent research indicates that NAD+ plays a key role in the creation and activation of the sirutins.

Doctors out of the Department of Developmental Biology at the Washington University School of Medicine were the first to show this link in 2014.2 The authors note:

NAD(+) levels decline during the aging process and may be an Achilles’ heel, causing defects in nuclear and mitochondrial functions and resulting in many age-associated pathologies. Restoring NAD(+) by supplementing NAD(+) intermediates can dramatically ameliorate these age-associated functional defects, counteracting many diseases of aging, including neurodegenerative diseases.”

These findings were corroborated in 2016 when the scientific journal Rejuvenation published an article where the authors discussed how low NAD+ levels are DIRECTLY associated with cellular aging, and they emphasized that this process can be prevented by increasing the levels of NAD+ within cells.

So is NAD+ a fountain of youth? It could be. More research needs to be done. But one thing seems clear: It certainly has an impact on your body at the cellular level, and it may just turn back the hands of time.

NAD+’s Role in Exercise and Endurance

It is well known that ATP levels get depleted in muscles while exercising. This simply stands to reason. However, until recently we didn’t know how NAD+ was implicated in this process.

A study done in 2010 using both trained and untrained healthy volunteers helped tease out the relationship. The researchers showed that intense exercise decreases NAD+. When the study participants took an antioxidant supplement containing pycgnogenol (which stimulates NAD+ production and protects it from turning into the inactive, oxidized form of the molecule), NAD+ levels increased and exercise performance and “time to fatigue” also improved.3

Using a direct IV infusion of NAD+ is certainly a more efficient way of raising NAD+ levels than taking an indirect, oral supplement that acts as a stimulator for NAD+. So we know that this IV can improve athletic performance, and it likely has a more pronounced effect than the study cited above.

NAD+’s Role in Reversing Chronic Illness and Neurodegenerative Disease

At the end of the day, health and energy production are intimately related. If your mitochondria aren’t functioning or they are producing too little energy, you are going to experience symptoms. When the problem becomes too severe it turns into illness.

The link between NAD+, energy production, and chronic illness really couldn’t be clearer. Low NAD+ results in low ATP levels. Low ATP levels quickly depletes your cell’s energy reserves. Left unchecked, and this can lead to cell death.

How all of this shows up in your body depends on which cells are being affected. If nerve cells are affected, the lack of energy may manifest itself as depression, anxiety, fatigue and lack of focus. In severe cases where the nerve cells are dramatically impacted or even die, neurodegenerative illness like Parkinson’s disease and Multiple Sclerosis may manifest themselves. In the worst cases—like mass cellular death—this may lead not only transient symptoms such as tremors, spasms, tingling, numbness and blurred vision, but to permanent, irreversible disability.

If your heart cells are affected, it can lead to cardiovascular disease. If your lung cells are affected pulmonary disorders may present themselves. Basically, when the cells in your organs and tissues don’t have enough energy, illness results.

IV NAD+ combined with IV amino acids can be beneficial in restoring energy production—especially in the nerve tissue—and may even prevent permanent damage.

One 2014 study showed that rat nerve cells pre-treated with high concentrations of NAD+ before being deprived of oxygen survived the challenge and recovered much faster than cells not bathed in the extra protective NAD+.4

A more recent review article published in 2015 builds on this. After scientists examined all of the available scientific evidence they recommended NAD+ supplementation and restoration as a new therapeutic opportunity for a wide variety of neurodegenerative diseases.5

NAD+’s Role in Addiction:

Perhaps the most exciting and controversial use medicine has found for NAD+ so far is in the treatment of drug and alcohol dependency. Despite the controversy, the basic science is sound, and the early results are extraordinary. Patients are reporting total recovery from addiction within 7-14 days with no subsequent craving for their substance of choice and few withdrawal symptoms.

To understand NAD+’s relationship to drug and alcohol abuse, let’s look at its role in the metabolism of alcohol

After you drink, your body tries to process and eliminate every molecule of alcohol. This is done in the liver where ethyl alcohol is converted to acetaldehyde. NAD+ is critical to this process. Alcohol donates one hydrogen molecule IRREVERSIBLY to NAD+ converting it to NADH. That NAD+ molecule is now gone and the NADH in your system begins to build. As this happens repeatedly, the NAD+/NADH balance swings out of favor, and serious problems can occur.

In a 2012 review article, the authors conclude that it is this NAD+ depletion and NADH accumulation that disables the mitochondria in your liver cells from being able to make energy ATP.6 This leads to liver cell death. Your body is then even less capable of processing the alcohol out of your system, which may set the stage for chemical dependency.

Building on this principle, several addiction clinics in the United States have been using IV NAD+ combined with IV amino acid therapy with great success. Dr. Ken Starr of the Wellness Group in San Louis Obispo and Dr. Metsayer of the Springfield Wellness Clinic in Louisana have been using a combination of daily IV infusions of NAD+ and amino acids for treatments lasting 10-14 consecutive days to drastically alleviate the symptoms of withdrawal from opiates, benzodiazepines and alcohol.

Their therapy, termed “Brain Restoration” (BR+) is many times combined with traditional FDA approved medications such as suboxone and naltrexone to achieve a much smoother transition to sobriety with drastically fewer unpleasant symptoms. Remarkably, many of their patients report the subjective loss of “craving” for the drug previously abused after the NAD+ and amino acid combination therapy.

How I Reversed My Parkinson’s Disease Symptoms

What is Parkinson’s Disease?

Parkinson’s disease is a neurological condition afflicting about 1 percent of men and women over the age of seventy years old. Individuals with Parkinson’s disease have tremor of the hands, rigidity, poor balance, and mild intellectual deterioration. The tremor is most apparent at rest and is less severe with movement. Shaking or tremor is an early symptom of Parkinson’s disease. Progression leads to trembling in arms, legs, jaw, and rigidity or stiffness of the limbs.

Research has shown that Parkinson’s disease occurs when a small region in the brain, called the substantia nigra, begins to deteriorate. The neurons of the substantia nigra use the brain chemical dopamine. Tremors begin to happen and movement slows when you have inhibited dopamine function. Dopamine is broken down in the brain by an enzyme called monoamine oxidase (MAO). When the activity of MAO is inhibited, dopamine stays around longer and this benefits those with Parkinson’s disease.

What Helps Parkinson’s Disease?

There is currently no official cure for Parkinson’s. As the disease progresses, patients often suffer from cognitive decline. Existing treatments can help ease symptoms, but over time the drugs themselves can cause debilitating side effects.

Carbidopa is most commonly used for Parkinson’s disease. However there can be a number of negative side effects and problems associated with the administration of carbidopa during treatment of Parkinson’s disease. This is thought to be because the administration of carbidopa will deplete the body of vitamin B-6, l-tyrosine, l-tryptophan, 5-hydroxytryptophan (5-HTP), serotonin, and sulfur amino acids. The properly balanced administration of mucuna pruriens (natural form of l-dopa), and not carbidopa, in conjunction with 5-HTP, l-tyrosine, l-cysteine and other supportive nutritional supplements can improve benefits.

A research study, led by Dr. Marty Hinz and printed in the International Journal of Internal Medicine was done with 254 Parkinson’s patients who were newly diagnosed, with no previous treatment, and those who were diagnosed more than 20 years before and had tried many other medical treatment options. It showed that these supplements were helpful in the treatment of Parkinson’s Disease.

In additional studies, Dr. Hinz has pioneered new research showing that replacing carbidopa with mucuna puriens can arrest symptoms of Parkinson’s at any stage and in many cases reverse it. Without carbidopa, he recommends a specific protocol of natural supplements similar to the protocol that I used to reverse my Parkinson’s. My personal protocol is listed below, however, it should not be attempted if one is taking carbidopa.

A clinical study of 885 Parkinson’s patients validated that in 78% of the cases, patients taking 10mg per day of NADH reported positive improvements in their condition. From my personal experience and in helping hundreds of people at my wellness retreats in Northern California, the following suggestions have helped all brain disorders.

These suggestions for optimal brain function should not be followed if you are currently taking prescribed drugs for Parkinson’s. Researchers at the Buck Institute for Research on Aging are continuing their investigation into the potential usefulness of lithium in the treatment of Parkinson’s disease.

New research by Buck scientists published online in Brain Research shows that low doses of lithium administered to laboratory mice reduced involuntary motor movements, a troubling side effect of the medication most commonly used to treat Parkinson’s disease.

Prior to the lithium dosing, the mice were given a drug — sold under the name Sinemet — that is used to boost levels of the neurotransmitter dopamine, which is lost in Parkinson’s disease. While Sinemet is the most effective treatment for managing Parkinson’s disease symptoms, long-term use causes abnormal involuntary movements.

“For patients, these side effects are just as devastating as the freezing that is associated with PD,” said senior Buck scientist Julie Andersen, who oversees the lab doing the lithium research, in a statement. “In our mice we saw significant behavioral improvement.”

Andersen’s research has focused on lithium as a potential treatment for Parkinson’s disease as well as its effectiveness when combined with drugs used to control the symptoms of the disease. Two previous studies conducted by Anderson’s lab found that low doses of lithium prevented symptoms of Parkinson’s disease in mice genetically engineered to develop the disease.

“We clearly saw a prevention of the motor difficulties we would expect to see in the animals,” Andersen said in a release. “The treatment also protected the area of the brain that is normally damaged by Parkinson’s.”

Unlike most medications, lithium is a naturally occurring element, not a developed molecule. The U.S. Food and Drug Administration approved its use for treating bipolar disorder in 1970, and it has proven effective for treating mood disorders and suicidal thoughts. Studies have suggested that low doses of lithium also have a protective effect in other neurodegenerative diseases including Alzheimer’s and Huntington’s.

In the new study, mice received a dose of lithium equivalent to about a quarter of what humans receive for treatment of psychiatric diseases. That is important because high doses of lithium have been linked to hyperthyroidism and kidney damage.

The Parkinson’s Study Group, a not-for-profit network of Parkinson’s centers, has expressed interest in initiating a clinical trial to assess the efficacy of low doses of lithium in treating Parkinson’s disease, said Buck Institute spokeswoman Kris Rebillot.

Low-dose lithium reduces side effects from most common treatment for Parkinson’s disease

Low-dose lithium reduced involuntary motor movements — the troubling side effect of the medication most commonly used to treat Parkinson’s disease (PD) — in a mouse model of the condition that is diagnosed in about 60,000 Americans each year. The third in a series of studies from the Andersen lab involving PD and low-dose lithium, the results add to mounting evidence that low-doses of the psychotropic drug could benefit patients suffering from the incurable, degenerative condition.

This study, published online in Brain Research, involved Parkinsonian mice that were given Carbidopa/Levodopa (sold as Sinemet®), a drug used to boost levels of the neurotransmitter dopamine, which is lost in PD. While the medication remains the single most effective agent in the management of PD symptoms, long-term use causes its own side effects, among them abnormal involuntary movements or AIMS. Buck professor and senior scientist Julie Andersen, PhD, says AIMS become problematic for 30 percent of patients after four to six years of treatment with Sinemet, with 90 percent of patients suffering from the complication after nine years of chronic use. “For patients these side effects are just as devastating as the freezing that is associated with PD.” “In our mice we saw significant behavioral improvement.”

In this study, Andersen and her team dosed the mice with an amount of lithium equivalent to about a quarter of what humans receive for the treatment of psychiatric diseases. Researchers found that lithium boosted the expression of tyrosine hydroxylase which increases dopamine synthesis via the inhibition of calpain-1, an enzyme that normally reduces dopamine synthesis.

In earlier studies, Andersen’s team found that low-dose lithium was protective in two different mouse models of PD. Treatment in mice with a human mutation for PD began when the animals reached late middle-age, the human equivalent of about 60, which is the average age of onset of Parkinson’s in humans. “We clearly saw a prevention of the motor difficulties we would expect to see in the animals,” said Andersen. “The treatment also protected the area of the brain that is normally damaged by Parkinson’s.”

Plans for a clinical trial of low-dose lithium for PD patients are in early stages. “This study suggests potential therapeutic benefit in PD,” said David K. Simon, MD, PhD, Associate Professor of Neurology at Harvard Medical School in Boston. Simon chairs the Scientific Review Committee for the Parkinson’s Study Group, a not-for-profit network of Parkinson’s Centers. “One caveat is that other agents that have shown clear efficacy in this model of PD have subsequently failed to show benefit in clinical studies in PD (e.g. CoQ10, creatine, and pioglitazone). However, this study provides additional evidence on top of prior work from Dr. Andersen’s lab and others that lithium may have therapeutic potential in PD, which is a hypothesis that should be tested in clinical trials,” he said.

Lithium is a naturally occurring element, not a ‘developed’ molecule like most medications. It was approved by the FDA for the treatment of bipolar disorder in 1970 and has shown to be effective for treating mood disorders and suicidal thoughts. Previous studies suggest that at low doses lithium has a protective effect in other neurodegenerative diseases including Alzheimer’s and Huntington’s.

Dr. David Simon, a professor of neurology at Harvard Medical School who chairs the Parkinson’s Study Group’s scientific review committee, said lithium’s effectiveness in treating Parkinson’s disease should be tested in clinical trials even though other agents that produced encouraging results in mice failed to show benefit in clinical studies.

The Clinical Testing:

NADH a successful treatment for ParkinsonsSince the mid-eighties, NADH has been successfully used to treat more than 3,000 Parkinson’s disease patients. These treatments were in Europe, and not in the US. A majority of the Parkinson’s disease patients experienced varying levels of positive improvements in their condition. To validate these results, an open label trial with 885 Parkinsion’s disease patients were given 25mg of NADH daily. About half of the patients received NADH by intravenous means (a needle), and the other half received oral tablets. The patients that received the 25mg of NADH oral tablets experienced the best results. (The actual Parkinson’s disease clinical study is available, see the link below.) The clinical study document is technical in nature, difficult to read, and is usually read by a healthcare professionals. We recommend taking the study to your healthcare professional and discuss its results.

The study found there were three things that had significant influence on the improvement in condition. They are:

  • the degree of the patient’s disability before the treatment
  • the duration of the disease within the patient before the treatment
  • the age of the patient

In the simplest of terms, the younger patients, with a shorter duration of the disease, have a better chance to obtain the most significant improvements in Parkinson’s disease symptoms. The stages of the disease are important. If the Parkinson’s disease is in the most advanced stages, it is very difficult to show improvement.

The clinical tests showed 78% of the patients taking NADH experienced varying levels of beneficial improvement. The percentages in improvement were:

  • 19.3% of the patients showed a very good (30% to 50%) improvement
  • 58.8% of the patients showed a moderate (10 – 30 %) improvement.
  • 21.8 % of the patients did not respond to the NADH

Parkinson’s disease: The Illness

It develops gradually. As the disease progresses, it produces body tremors, a shuffling gait, and eventually a possible incapacity. Emotions may be affected. Mental capacity may be impaired. Depression often accompanies the disease. In the early stages of Parkinson’s disease, the face may show little or no expression or the arms may not swing when you walk. Speech may become soft or slurred. Parkinson’s disease symptoms worsen as the condition progresses over time.

The cause of Parkinson’s disease is unknown, but several factors appear to play a role, including:

  • The person’s genes. Researchers have identified specific genetic mutations that can cause Parkinson’s disease. These are uncommon except in rare cases when many family members are being affected by Parkinson’s disease.
  • Certain gene variations appear to increase the risk of Parkinson’s disease but it is a relatively small risk for each of these genetic variations.
  • Environmental triggers. Exposure to certain toxins, herbicides, pesticides and other environmental factors may increase the risk of Parkinson’s disease, but the risk is relatively small.
  • In summary, more research needs to be done to identify the factors that cause Parkinson’s disease.

In Parkinson’s disease, certain nerve cells (neurons) in the brain gradually break down and die. Many of the symptoms are due to loss of brain cells (neurons) that produce a chemical messenger in your brain called dopamine (also known as neurotransmitters). When the brain cells that produce dopamine, stop producing dopamine, the dopamine levels decrease, and that causes abnormal brain activity, which leads to signs of Parkinson’s disease. Parkinson’s disease is named for the English surgeon James Parkinson, who first described it in 1817.

 

Parkinson’s disease: Who Will Be Affected?

Who will be affected by Parkinson’s disease is unknown, but the following risks have been identified:

  • Age. Young adults rarely experience Parkinson’s disease. It ordinarily begins in middle or late life, and the risk increases with age. People usually develop the disease around age 60 or older
  • Heredity. Having a close relative with Parkinson’s disease increases the chances that you’ll develop the disease. However, your risks are still small unless you have many relatives in your family with Parkinson’s disease
  • Sex. Men are more likely to develop Parkinson’s disease than are women
  • Exposure to toxins. Ongoing exposure to herbicides and pesticides may put a person at a slightly increased risk of Parkinson’s disease

Parkinson’s disease: Diagnosis & Current Treatment:

It should be known that Dr. G. In occasional cases, the doctor may suggest surgery to regulate certain regions of your brain and improve the symptoms. Parkinson’s disease is treated with these drugs: sinemet, deprenyl (selegiline), L-DOPA (given with carbidopa to reduce side effects), amantadine and others.     ATP Life Energy - It is life's powerEvery living thing contains the chemical,adenosine triphosphate(ATP). ATP is used to provide energy for heat, nerve electricity, light (as in fireflies), and muscle movement. An ATP molecule is held together by strong electrical forces which are set free when the molecule is broken apart in a chemical reaction. Somehow these forces are converted into the kind of mechanical energy that will move our muscles. The part broken from the ATP molecule is a smaller molecule calledphosphate. After the phosphate is broken off, living organisms can put it back and use the ATP molecule again. To do this, a rebuilding chemical calledphosphagen, is used for a while until the powerful glycogen cycle takes over the production of fresh ATP energy. The glycogen cycle is a series of chemical steps in which sugar and other substances are used to make fresh ATP in large quantities. The whole process is so intricate that dozens of chemical reactions take place, literally, in the wink of an eye!

     

    Fuel for the Muscle Motors

    Muscular contraction is one of the most wonderful things in the operation of our body machinery. And though much about it remains a mystery, biochemists made many strides towards the understanding of this operation.In whatever form sugars are eaten, whether as starch, syrups, candy, or common table sugar, they are broken down by the juices of thealimentary canal(the digestive tract) to simple sugars. All these sugars are then changed to the simplest sugar,glucose, which is the only sugar found circulating in the blood.

    The amount of glucose in the blood stays remarkably the same. It increases in the disease known as diabetes, otherwise the sugar content of the blood is about the same in all healthy persons.

    The body’s heat comes mostly from the burning of glucose. Cold-blooded animals such as the frog have less sugar in their blood than we do. Birds, which are warmer than humans, have more.

    If we could not store so vital a substance as glucose in our bodies, we would need to eat constantly to maintain a steady supply of fuel. That would be a dangerous existence. If we were to fall asleep, we would never wake. Without food, our body’s machinery would grind to a halt. On the other hand, large amounts of free glucose cannot be kept in the body either. It is too readily used up. Our bodies can, however, store the glucose in a more stable, less readily used form –glycogen. Glycogen is made up of many glucose molecules that are hooked together.

    How Energy is Stored in the Body

There are large deposits of glycogen in the liver, in the muscles, and in the kidneys. As the body needs glucose, glycogen is broken down into glucose molecules to supply the body’s immediate needs. If there is any excess glucose in the blood, as there is after a meal, it is shipped to the glycogen storage depots.

    When energy is needed, glucose molecules are broken down to release energy in the form of heat. The release of energy takes place in an astounding series of reactions brought about within the body by chemical substances known asenzymes and co-enzymes.

    The earliest glimpses of these reactions were obtained, oddly enough, from studies of yeast cells, not of animals. Yeast cells, up to a point, use sugars for their energy exactly as we do.

    Before we can be qualified as engineers for the human body, this most marvelous of machines, we need to knowhow energy is releasedin chemical reactions.

    All forms of energy are interchangeable. That is, heat can be changed into motion; motion can be changed into electricity, which, in turn, can give light or heat again. Such conversions or changes are often very wasteful. The average steam locomotive loses over 90 percent of the energy of the steam as it converts the remaining 10 percent to motion. The average light bulb uses 90 percent of its energy in creating heat, the rest of the energy is used to produce light

     

     

    How Energy is Released

    How Energy is ReleasedEnergy cannot be destroyed. We can take a pound of coal and burn it in an ample supply of air while it changes to carbon dioxide. An amount of heat (n) will be given off. If we burn another pound of coal in a limited supply of air it will form carbon monoxide, but only about one-fourth (1/4) as much heat will be liberated as before. If we now burn all of this carbon monoxide to carbon dioxide, we get the rest of the original amount of heat (n). Whether we release the energy in one step or in several steps, the overall amount is the same.

    What about the energy in glucose? Measurements of energy are always based on the chemist’s unit weight, or molecular weight. Glucose weighs 180 grams. (One molecule of glucose weighs 180 times as much as one atom of hydrogen.) When a green plant makes glucose from carbon dioxide and water, it packs energy into it. Into about 180 grams (or six ounces) of glucose are packed 700 calories of energy. The calorie is a measure of heat energy. One hundred calories will heat one liter (about a quart) of ice-cold water to boiling. If we burn the 180 grams of glucose, 700 calories of heat will be released.

    Where are these 700 calories hidden? They are used to form the bonds that hold together the six carbon atoms, the twelve hydrogen atoms, and the six oxygen atoms that make up the one glucose molecule. There is energy in chemical bonds.(1 glucose = C6 H12 O6)

    Imagine a dozen large springs from a mattress squeezed into a hatbox. A good deal of energy had to be used to squeeze the springs together before the box lid could be safely locked. If the lid is opened, the jumping springs will release the same amount of energy that was used to put them into the box. This is a fair comparison of the energy used to lash the atoms of carbon, hydrogen, and oxygen together to form glucose. (This bond energy has nothing to do with the energy within the nucleus of the atom, known as atomic energy. And, of course, bond energy is not nearly so great as the monstrous energy of the nucleus.)

    Glucose is Changed into Energy

Some bonds have more energy packed into them than others. The cell gets its warmth and its energy for work from the breaking of the bonds of glucose molecules. Cells have a complicated set ofenzymes and co-enzymesfor the step-by-step whittling of bonds of the glucose molecule and the step-by-step release of energy from these bonds. NADH is the coenzyme that plays the primary role in the release of energy.

    Some of these steps are well known, and the overall picture is clear. The bonds in glucose are broken one at a time, and phosphates (salts containing phosphorus) play an important role in the process.

    Phosphate molecules store much of the released energy in a form more useful to the cell. The howling wind has a lot of energy. The farmer’s windmill catches some of that energy and at once puts it to work pumping water from a well. But some of that energy is also stored by the charging of batteries. The wind cannot light up the farmer’s house, but the battery can. The heat from a crumbling glucose molecule does not make our legs move. It is the energy in the phosphate bond that supplies the energy that makes the muscles move. Phosphate bonds are our batteries. They are the stored energy for life’s every need.

    How does it work, this marvelous battery to which we owe our lives?  The battery is a molecule – a molecule calledadenosine triphosphate– abbreviated asATP. Onto one molecule of ATP are lashed two, special phosphate groups. Ten calories of energy are packed into each of those bonds which hold these phosphates to the ATP. These phosphate cementing calories are the main form of energy the cell can use for its many tasks.

     

    How Stored Phosphate-Bond Energy is Used in Our Body’s Muscles

    This stored phosphate-bond energy is used in a remarkable manner. Suppose a muscle cell needs ten calories of energy to set off a muscle contraction. A unit of ATP is alerted to act by an agent, called a coenzyme, which splits off one phosphate unit and then delivers the required ten calories. In the process, the ATP is reduced to ADP (adenosine diphosphate). If the energy set free by the splitting off of phosphate units is not used for muscle contraction, it can be transferred to the other compounds in the muscle such as creatine and glucose. In so doing, the glucose is taken from a glycogen storage depot and begins to be broken down. The energy flowing from the crumbling glucose is used to furnish heat, convert some of the broken-down glucose back to glucose, and attach the liberated phosphate units to ADP to reform ATP. When this cycle is completed, the ATP units are ready for any new emergency.

    The ready supply of the stored ATP energy for movement is marvelously useful to all animals in emergencies. Sometimes an animal needs enormous amounts of energy when there is not enough time to break down glucose. After a vigorous sprint to the bus a person may pant for minutes before they can settle down to the calm reading of the morning paper. The panting is the body’s way of taking in large amounts of oxygen needed for the burning of glucose to build back the ATP used up by the exertion of hurrying to the bus.

    How Phosphate-Bond Energy Moves Our Legs

    We are Sun MachinesThe bones in our legs are moved by the muscles attached to them. These musclesalways come in matched pairs. As one muscle tightens or contracts, its opposite relaxes; then the other one contracts, and the first one stretches or relaxes. Our arms, legs, and fingers move by means of this alternate relaxing and contracting of muscles. The energy for this work is provided by the phosphate-bond energy of ATP. The energy for the replacement of this phosphate bond the sun’s energycomes from glucose, and this energy in turn comes from the sun. So we are sun machines, fantastically complex sun machines, but sun machines nonetheless.

     

     

    A Look Inside the Muscle Machinery

    A look inside your muscle fibers you find ATPLet us lift the hood and take a look at the machinery. Muscle cells are made up of many long, tiny fibers composed chiefly of a protein calledmyosin.These fibers are chock full of ATP. In the early 1950’s Albert von Szent Gyorgyi, a Hungarian born biochemist, (who had come to the United States,) brought about muscular contraction in a test tube. In 1937 he received the Nobel Prize in Medicine for his earlier work. He managed to cause the myosin threads of muscle to lose all of their ATP. He then added to these lengthened threads, some ATP. The long threads curled up instantly on contact with the source of energy. Contraction of the myosin threads is the mechanism for contraction of a muscle. This demonstration is a milestone in the history of science, for in Szent-Gyorgyi’s words

    Clinical Testing: Sleep Deprivation Study

    How many hours of sleep do we needA NADH nutritional supplement was clinically tested in a Sleep Deprivation Study. NADH was found to improve the cognitive functioning or “thinking performance” following 24 hours of sleep deprivation. Sleep deprivation is a common problem affecting most people sometime in their life. It impacts otherwise healthy individuals who cross time zones, work during evening hours, the night shift or late nighttime hours, or have infant children, or students who stay up all night studying. It impacts patients with sleep disorders, certain psychiatric disorders, and medical conditions such as those that produce chronic pain. Sleep deprivation leads to a decline in cognitive performance (a thinking ability), impacting the quality of waking time and, if severe enough, can lead to vehicle collisions and other negative occupational consequences (work related mistakes or accidents).

    Dr Birkmayer has used NADH to help CFS patients increase energy and alertness.  He used NADH to help people reduce the effects of jet lag.  Dr. Birkmayer has used NADH to help people with the symptoms of sleepiness caused by sleep deprivation.  Dr. Birkmayer wanted to prove his results under a carefully run clinically trial.  His company funded a sleep deprivation clinical study to prove NADH’s effectiveness in rigorous testing and under the watchful eyes of a research hospital.

    In White Plains, N.Y. Researchers in the Sleep-Wake Disorders Center of N.Y. Weill Cornell Medical Center, clinically tested (stabilized) NADH to improve alertness, mood, and performance on cognitive or thinking ability tasks.  There were 25 healthy middle-aged adults tested. The tests were performed after one night of total sleep deprivation.

    The NADH Sleep Deprivation study was a double-blind, crossover study. In double-blind studies, the patients, the doctors, the nurses, and the research administrators monitoring the results DO NOT KNOW which patients received the NADH and which patients received the placebo (the sugar pill).

    The clinical study results proved the NADH recipient subjects performed significantly better on some measures of cognitive performance following one night of total sleep deprivation (up for 24 hrs) when compared to those subjects taking the placebo (the sugar pill).  One hour after consuming 20mg of NADH, the overall performance efficiency (number of correct answers per minute) for the subjects taking NADH was significantly higher than for those subjects taking a placebo.  In a second analysis, after taking NADH, math throughput and visual sequence comparison speed were also significantly better for those subjects taking the NADH.  All 25 subjects provided self-reported alertness, feeling of sleepiness , and mood.  The self-reported and feeling of sleepiness were objectively quantified.  All 25 subjects (both groups, those taking NADH and those taking the placebo) DID NOT differ in their self reporting comments.  Although several subjects reported the typical effects of sleep deprivation, no adverse effects were attributed to NADH.   Yet the group taking the NADH performed significantly better in testing.

    This study is among the first to rigorously evaluate a non-prescription substance other than stimulants (like coffee or other caffeine drinks) for alleviating the effects of sleep deprivation. Dr. Margaret Moline, the Director (and Lead Researcher) of the Sleep-Wake Disorders Center at the Westchester Division of New York-Presbyterian Hospital in White Plains NY, states, “NADH is the first non-stimulant, non-herbal product to show signs of improved cognitive performance, despite normally reported increased sleepiness and fatigue following sleep deprivation. These results suggest that NADH may have an important role to play in mitigating some in the unavoidable effects of sleep deprivation.”

Investigators using structural MRI have reported diminished cortical gray matter, most prominently in the prefrontal cortex (PFC), in alcoholic patients in treatment (Pfefferbaum et al., 1998). In another study, researchers showed that alcohol-dependent individuals had reduced whole brain, prefrontal cortical, and parietal cortical gray matter compared with controls (Fein et al., 2002).

L-Trytophan is the precursor (building block) for serotonin and melatonin. Melatonin helps to regulate sleeping and waking. Serotonin is one of the major inhibitory neurotransmitters in our brain, deficiencies of which can cause symptoms like nervousness, anxiety, and insomnia. It would be very convenient if one could simply correct neurotransmitter imbalances with specific dietary changes. Unfortunately, even though many foods contain amino acids, it is not possible to design a diet where a person can obtain enough amino acids in the correct proportions to significantly increase specific neurotransmitter levels over time. Believe me, we’ve tried.

Why Diet Isn’t Enough

Diet alone can’t make up for specific neurotransmitter imbalances because the uptake of amino acids by the body is not selective: foods contain an array of amino acids and no food contains just the precursors needed to specifically affect only serotonin and/or dopamine. Therefore, when you eat food, your body takes up many different amino acids at random, making it impossible to target just the neurotransmitters we are trying to affect. This is where supplementation can play a pivotal role.

Amino Acid Therapy Can Help Drugs Work Better

Amino acid precursors of serotonin and dopamine have two primary applications. First, proper use of amino acid precursors will keep drugs that work with neurotransmitters from depleting neurotransmitters, thus allowing the drugs to keep functioning as intended. This is the situation when someone is taking a reuptake inhibitor that ‘stops working’. When we provide such a person with balanced amino acid therapy, we give the body what it needs to make the necessary neurotransmitters, which increases neurotransmitter levels. Drugs that work by shuffling around neurotransmitters (such as SSRI, SNRI and other medications) have more neurotransmitter to work with under this scenario so they can appear to start ‘working’ again. In this way, a person can continue to use the medication without fear of further neurotransmitter depletion due to the drug. In this case using amino acid therapy helps the drug do its job more effectively and efficiently.

Amino Acid Therapy Can Be Used to Eliminate the Need For Certain Medications

Second, proper use of amino acids can also serve as a stand-alone method to manage disorders related to neurotransmitter imbalance. Many people, for one reason or another cannot or will not take certain medications. Most disorders are not due to a drug deficiency; they are due to a nutrient deficiency or improper neurotransmitter function. By supplying the body the nutrients it needs to produce the correct balance of neurotransmitters, we can often restore proper function without the need for medication. In this case, amino acid therapy can provide a viable option to restore optimal neurotransmitter function, effectively eliminating symptoms without the need for medication.

With all of these ways to throw your neurotransmitter levels out of balance, you may think it would be impossible to address them all. Luckily this isn’t so.

There are really only two major effects that all of the causes of neurotransmitter imbalance listed above have on the body. They either (1) cause a decrease in the amount of neurotransmitter available or (2) they cause damage to the post-synaptic neuron. In the first case, restoring proper neurotransmitter levels using amino acid therapy can quickly and effectively eliminate the relative deficiency and alleviate symptoms. In the second, more directed therapy, often requiring specific testing can make up for the damage and restore proper function.

Overcoming Neuronal Damage

Neurons are intended to function for life. Loss of a neuron to cell death is permanent. As damage to the receptors and neurons progress the neuron’s function slowly fades until neurotransmitter production decreases and eventually stops over time.

A nerve is made up of many neurons, often called a nerve bundle. As individual neurons fade and/or die, the amount of neurotransmitter sent through a nerve bundle decreases, as depicted here:

As this process continues, a person will experience more and more symptoms of imbalance; these can include mood swings, headaches/migraines, OCD tendencies, decreased focus/concentration, decreased memory, anxiety, sleep imbalances, hormone imbalances, food cravings and increased pain amongst other symptoms.

The Solution: Turn Up the Voltage

As we discussed earlier, the only way to increase neurotransmitter levels in the brain is to take amino acids (along with the necessary cofactors) which can cross the blood brain barrier where they can then be made into neurotransmitters. Increasing neurotransmitter levels through the neurons is analogous to increasing the voltage in an electrical wire, where by turning up the voltage you get more electricity out the other end of the wire.

It may help to think of it like this: when neurotransmitter disorders are present, some of the neurons within the nerve bundles are damaged to the point that the electricity flowing out of them is diminished to the point where symptoms develop. Increasing neurotransmitter levels through the administration of properly balanced amino acid therapy will effectively increase the voltage in the remaining viable neurons in the bundle. This causes the electrical flow out of the damaged nerve bundles to increase to the point that normal regulation and/or control is once again observed. In this state, from a clinical standpoint, the symptoms of disorder are under control.

If this all sounds like Charlie Brown’s teacher (i.e., “Wah, wa, Wah…”) try this: There isn’t a feedback loop in the brain for neurotransmitter production. Therefore, even if 50% of the neurons in a nerve bundle are damaged or destroyed, if we give the remaining neurons more amino acids (in the right dosages), the remaining healthy neurons will manufacture more neurotransmitters, effectively alleviating symptoms and restoring normal function.

In essence, we give the healthy neurons what they need to make up for the damaged ones; when we get the balance right, it’s as though the damaged neurons don’t exist and you function normally.

Neurotransmitters are a class of chemical messengers in the body that help regulate, either directly or indirectly, most of the other systems and functions in the body. This is often referred to as ‘neurotransmitter testing’ although that term does not properly describe the extent of the test.  As we discussed earlier, neurotransmitters are chemical messengers; when neurotransmitter levels are too low or imbalanced, the messages get garbled and symptoms of disease or disorders develop. The way to correct this is by restoring proper neurotransmitter balance by providing the body with the nutrients it needs to restore proper neurotransmitter function. However, it isn’t quite as simple as giving the body the nutrients and having it ‘make’ the necessary neurotransmitters.

Transporters, which are molecules in the cell walls, facilitate the movement of neurotransmitters in and out of the cells, including the neurons.

Neurotransmitters cannot cross the walls of the neurons on their own as they are water-soluble and the cell walls are made of fat. Water and oil (fat) do not mix. The transporters remedy this by escorting the neurotransmitters in and out of the neurons.

When there is damage to the neurons, a signal is sent throughout the body to all the transporters that tells them to alter the flow or neurotransmitters in a way to try and compensate for the problem. When the damage is minor, the transporters can compensate for neurotransmitter imbalances by utilizing nutrients from a healthy diet. However, when significant damage occurs, properly balanced amino acid therapy needs to be used to correct the problem.

Organic Cation Transporter Optimization

A special transporter called the ‘organic cation transporter (OCT) is primarily responsible for the transport of serotonin and the catecholamines (dopamine, norepinephrine, and epinephrine) as well as their amino acid precursors across the cell wall. These transporters are found throughout the body in many tissues, including the kidneys, gut, liver as well as the brain and function similarly in all tissues throughout the body. This means that if we affect change to the OCTs with amino acid therapy it affects all the OCTs throughout the entire body.  This is very important.

By giving the body properly balanced amino acid therapy we can define the exact amounts of neurotransmitters and amino acids needed to restore proper functioning of the OCTs in the kidneys. However, because this also affects the OCTs in all the other tissues, including the brain in exactly the same way, testing the urinary excretion of neurotransmitters while taking a known amount of balanced amino acids allows us to restore proper functioning within the brain. This restores the flow of electricity in the neurons and leads to the relief of symptoms.

So what we are really doing is using amino acid therapy to help optimize the function of the OCTs in the neurons to facilitate proper signaling. (11) This is akin to unscrambling the messages so that the brain gets the proper signals. When the signals are no longer crossed, your body sends the right signals, your symptoms disappear and you get your life back.

Organic Cation Transporter (OCT) Assay Interpretation is a mouthful, but it is the proper name of the “neurotransmitter test” that is clinical useful. Amino Acid Therapy and Parkinson’s Disease

Historically, the most effective way to manage Parkinson’s disease involved using L-dopa to increase dopamine levels in the brain. L-dopa can cross the blood-brain-barrier and is freely synthesized into dopamine if the necessary co-factors are present.

The cause of Parkinson’s disease is damaged post-synaptic dopamine neurons in the brain which cause a steady and dramatic decline in dopamine production and function; this results in the symptoms associated with Parkinson’s disease. L-dopa is required in order to raise dopamine levels high enough to alleviate symptoms.

However, L-dopa is associated with numerous side effects when given as a stand-alone therapy. Many of these side effects are due to the fact that the administration of L-Dopa alone may deplete L-tyrosine, L-tryptophan, 5-hydroxytriptophan (5-HTP), serotonin and sulfur amino acids.

This is exacerbated by the fact that Parkinson’s disease itself is associated with a depletion of tyrosine hydroxylase (the enzyme responsible for catalyzing the conversion of L-tyrosine into L-Dopa), dopamine, serotonin and norepinephrine.

Carbidopa is often given along with L-dopa to try and relieve some of the side effects of L-dopa administration. Carbidopa is a general decarboxylase inhibitor, which means it inhibits the peripheral synthesis of both serotonin and dopamine (as well as norepinephrine and epinephrine). Although this approach may relieve some of the side effects of L-dopa administration, especially nausea, it is associated with an incredibly long and severe list of side effects and has been implicated as being the causative agent in the progression of Parkinson’s disease.

The ONLY way to address the underlying neurotransmitter imbalances associated with Parkinson’s disease is to take the proper balance of L-dopa in conjunction with 5-HTP, L-tyrosine, L-cysteine and necessary cofactors to restore optimal neurotransmitter function. The protocol necessary to address each person’s unique underlying causes needs to be tailored to their exact needs. In addition, special attention needs to be placed on when a person takes their amino acids, as timing can be as important as the dosage itself in regards to the elimination of symptoms.

This can only be accomplished by working with a provider trained in the proper use of amino acid therapy and is imperative for long term success.
